Biological Background: CD177 Function and Localization
- CD177, also known as human neutrophil alloantigen 2a (HNA-2a), NB1 glycoprotein (NB1 GP), and polycythemia rubra vera protein 1 (PRV-1), is a GPI-anchored glycoprotein mainly expressed on neutrophils.
- In complex with beta-2 integrins (ITGAM/CD11b and ITGB2/CD18), CD177 mediates TNF-primed neutrophil activation, including degranulation and superoxide production. Related references: PMID:21193407
- CD177 modulates neutrophil trafficking by preventing beta-2 integrin internalization and attenuating chemokine signaling, thereby favoring adhesion over migration. Related references: PMID:28807980
- Through heterophilic interaction with PECAM1 on endothelial cells, CD177 contributes to neutrophil transendothelial migration in vitro, though in vivo studies suggest it is dispensable for bacterial infection-induced recruitment. Related references: PMID:17580308 PMID:23461681
- CD177 serves as a membrane receptor for mature protease PRTN3, enabling its display on the neutrophil surface and potentially enhancing endothelial junctional integrity during diapedesis. Related references: PMID:17244676 PMID:18462208 PMID:23202369
- Subcellularly, CD177 localizes to the cell membrane, membrane rafts, cytoplasmic granule membranes, and lamellipodia; it can also exist in a secreted form.
- Tissue expression is highly specific to the neutrophil lineage, beginning at the metamyelocyte stage, and is found on a subpopulation of mature neutrophils with variable percentage across individuals; it is overexpressed in granulocytes from polycythemia vera and essential thrombocythemia patients. Related references: PMID:10753836 PMID:17244676 PMID:18462208 PMID:24926686 PMID:12377969 PMID:12675722 PMID:17580308 PMID:21193407 PMID:27227454 PMID:28240246 PMID:28807980
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 22–407 of human CD177 (NP_065139.2), covering almost the entire extracellular domain; thus, the antibody is likely suitable for detecting native, cell surface CD177 by flow cytometry (FC) and ELISA.
- For flow cytometry applications, consider using a neutrophil-enriched population or whole blood samples from healthy donors, noting that CD177 expression is restricted to a subset of mature neutrophils and can vary between individuals.
- For ELISA, the antibody may be used as a capture or detection reagent; empirical optimization of working concentrations and buffer conditions is recommended.
- Given its role in TNF-primed neutrophil activation, the antibody could be employed to investigate CD177 expression and function in inflammatory contexts, but validation in the relevant sample system is essential.
- Since the protein exists as multiple isoforms (predicted molecular weights ~46 kDa full-length, with possible 27 kDa and 15 kDa forms), confirm the detected band sizes in your experimental setup.
