Biological Background: CD177 Function and Localization
- CD177 mediates activation of TNF-primed neutrophils, including degranulation and superoxide production, in association with beta-2 integrin ITGAM/CD11b and ITGB2/CD18. Related references: PMID:21193407
- By preventing beta-2 integrin internalization and attenuating chemokine signaling, CD177 favors neutrophil adhesion over migration. Related references: PMID:28807980
- CD177 heterophilic interaction with PECAM1 on endothelial cells facilitates neutrophil transendothelial migration in vitro, though it appears dispensable for in vivo neutrophil recruitment during bacterial infection. Related references: PMID:17580308, PMID:23461681
- Acts as a receptor for mature protease PRTN3, enabling its display on the neutrophil cell surface. Related references: PMID:17244676, PMID:18462208
- By displaying PRTN3, CD177 may enhance endothelial junctional integrity and vascular integrity during neutrophil diapedesis. Related references: PMID:23202369
- Subcellularly, CD177 localizes to the cell membrane, membrane rafts, secreted forms, cytoplasmic granule membranes, and lamellipodia.
- Highly expressed in normal bone marrow; during neutrophil differentiation, expression begins at the metamyelocyte stage. Expressed only in a subset of mature neutrophils, with percentage varying across individuals. Also upregulated in polycythemia vera and essential thrombocythemia granulocytes.

Experimental Guidance and Technical Tips
- The immunogen covers amino acids 35–285 of human CD177, encompassing the extracellular domain likely recognized in native and denatured forms. For WB, consider a reducing or non-reducing protocol as the protein contains disulfide bonds; the predicted molecular weight is 51 kDa, but glycosylation may shift the apparent band.
- For ELISA applications, consider pairing with a validated detection antibody reactive to a non-overlapping epitope, and include appropriate antigen controls.
- CD177 expression is restricted to a subset of neutrophils; validate reactivity using primary human neutrophils or suitable cell lines that endogenously or ectopically express CD177.
- The antibody cross-reacts with human and mouse CD177, enabling cross-species studies; perform internal validation for each species and application.
