  • Neural cell adhesion molecule L1 (L1CAM, also designated CD171) is a cell adhesion molecule of the immunoglobulin superfamily that exists in multiple isoforms via alternative splicing.
  • It mediates cell adhesion dynamics and generates transmembrane signals at tyrosine kinase receptors, playing essential roles in nervous system development and function.
  • During brain development, L1CAM is critical for neuronal migration, axonal growth and fasciculation, and synaptogenesis; in the adult brain, it contributes to synaptic plasticity and structural dynamics.
  • L1CAM localizes to the plasma membrane and is enriched at specialized neuronal projections, including growth cones, axons, and dendrites.
  • The protein is a glycophosphoprotein with immunoglobulin-like domains and disulfide bonds, and its phosphorylation regulates adhesive and signaling functions.

  • Immunogen design: The immunogen targets the intracellular C-terminal region (amino acids 1143-1257) of L1CAM, conserved across human, mouse, and rat, supporting the observed cross-reactivity. This region is not expected to be glycosylated, so the antibody may recognize both glycosylated and unglycosylated forms.
  • Western blotting: The predicted molecular weight is 140 kDa, but extensive glycosylation typically shifts the apparent band to ~200-220 kDa. Use reducing and denaturing conditions, and validate with brain or neuronal cell lysates.
  • Immunofluorescence/ICC: Permeabilization is required to detect the intracellular epitope. Fixation with 4% PFA and permeabilization with 0.1% Triton X-100 are suggested. Co-stain with neuronal markers to confirm specificity in neurons and projections.
