- Sialic acid-binding immunoglobulin-like lectin 5 (Siglec-5), also known as CD170, CD33L2, or OBBP2, is a member of the Siglec family of transmembrane receptors that recognize sialic acid-containing glycans.
- It functions as a putative adhesion molecule that mediates sialic acid-dependent binding to cells, with equal affinity for α-2,3-linked and α-2,6-linked sialic acids.
- The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface, modulating its binding activity.
- Siglec-5 is a membrane protein with an immunoglobulin-like domain and is predominantly expressed on monocytic and myeloid lineage cells, including peripheral blood monocytes and neutrophils.
- Tissue expression is high in peripheral blood leukocytes, spleen, and bone marrow, and lower in lymph node, lung, appendix, placenta, pancreas, and thymus.
- It contains disulfide bonds and is classified as a lectin and cell adhesion molecule, with structural features including Ig domains and a transmembrane helix.
