Biological Background: CD168/RHAMM Function and Localization
- Encoded by the HMMR gene, CD168/RHAMM (Hyaluronan Mediated Motility Receptor) is also known as Intracellular Hyaluronic Acid-Binding Protein (IHABP) and Receptor for Hyaluronan-Mediated Motility (RHAMM). The protein undergoes alternative splicing.
- Functions as a receptor for hyaluronic acid (HA), playing a key role in cell motility. Upon HA binding, it triggers phosphorylation of multiple proteins, including PTK2/FAK1, and regulates extracellular-regulated kinase (ERK) activity.
- Implicated in cellular transformation and metastasis formation, and may also act as a regulator of adipogenesis.
- Exhibits a dual subcellular distribution, localizing to the cell surface, cytoplasm, and the cytoskeleton, specifically associated with the spindle apparatus.
- Expressed in testis and breast tissue. Related references: PMID:22965910 PMID:8890751
- Classified under keywords such as glycoprotein, phosphoprotein, and hyaluronic acid-binding, indicating post-translational modifications and functional interactions.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 341-440 of human CD168/RHAMM (isoform based on NP_001136028.1). For detection of other isoforms or species variants, consider verifying epitope conservation.
- This polyclonal antibody is suitable for western blot (WB) and ELISA. The predicted molecular weight is 84 kDa; in WB, validate band size in relevant cell or tissue lysates, considering possible post-translational modifications.
- Cross-reactivity has been confirmed for human, mouse, and rat samples. For other species, perform sequence alignment or pilot testing to assess suitability.
- Since CD168/RHAMM is expressed in breast and testis, these tissues may serve as positive controls. Adjust sample preparation to account for its association with the cytoskeleton and spindle apparatus.
