Biological Background: CD166/ALCAM Function and Localization
- CD166/ALCAM (activated leukocyte cell adhesion molecule) is a transmembrane glycoprotein of the immunoglobulin superfamily, with multiple isoforms arising from alternative splicing, including a secreted variant that antagonizes membrane-bound functions.

Experimental Guidance and Technical Tips
- For Western blotting, use reducing SDS-PAGE conditions and an expected molecular weight of approximately 65 kDa; note that glycosylation may cause band shifts. Validate detection in lysates from ALCAM-expressing cells such as Jurkat or activated T-cells.
- When using this antibody in ELISA, consider optimizing coating concentrations and pairing with CD6-Fc or ALCAM-specific detection antibodies to establish a functional assay.
- Given the cross-reactivity with mouse, validation in mouse-derived samples is recommended to confirm species-specific binding and performance.
