Biological Background: CD166/ALCAM Function and Localization

  • CD166/ALCAM, also known as activated leukocyte cell adhesion molecule (ALCAM) or MEMD, is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily. It contains five extracellular Ig-like domains, a single transmembrane helix, and a short cytoplasmic tail.
  • CD166/ALCAM mediates both heterotypic cell-cell contacts via interaction with CD6 and homotypic interactions with ALCAM-expressing cells, playing critical roles in T-cell activation and proliferation. Related references: PMID:15048703, PMID:16352806, PMID:24945728
  • It contributes to the formation and maturation of the immunological synapse through its interaction with CD6, enhancing immune responses. Related references: PMID:15294938, PMID:16352806
  • Acts as a ligand for the inhibitory receptor LILRB4, thereby enhancing LILRB4-mediated inhibition of T-cell proliferation. Related references: PMID:29263213
  • Required for normal hematopoietic stem cell engraftment in the bone marrow and plays a role in hematopoiesis; required for maintenance of hematopoietic stem cell numbers. Related references: PMID:24740813
  • Mediates attachment of dendritic cells onto endothelial cells via homotypic interaction, and inhibits endothelial cell migration while promoting endothelial tube formation. Related references: PMID:23169771, PMID:15496415
  • Promotes neurite extension, axon growth, and axon guidance; retinal ganglion cell axons grow preferentially on ALCAM-coated surfaces.
  • Subcellular localization: primarily expressed on the cell membrane, but also found on cell projections like axons and dendrites; a secreted isoform exists. Detected on hematopoietic stem cells, lymphocytes, monocytes, dendritic cells, endothelial cells, and various tissues including spleen, placenta, and liver. Related references: PMID:24740813, PMID:23169771, PMID:15048703, PMID:16352806, PMID:9502422, PMID:7760007, PMID:15496415

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the entire extracellular domain (amino acids 28-526), ensuring recognition of the native protein. This makes the antibody suitable for detecting CD166/ALCAM in its native conformation in applications like IF/ICC and ELISA.
  • For Western blotting (WB), a band around 65 kDa is expected; consider using appropriate loading controls and optimize protein extraction methods for membrane proteins.
  • In IF/ICC, anticipate membrane and possibly punctate staining along cellular projections; validate with positive and negative cell lines (e.g., T-cells, melanoma lines).
  • For ELISA, pair with a suitable secondary antibody and validate assay conditions using recombinant protein standards.
