Biological Background: CD166/ALCAM Function and Localization

  • CD166 antigen (ALCAM, activated leukocyte cell adhesion molecule) is a transmembrane glycoprotein of the immunoglobulin superfamily. Alternative splicing yields membrane-bound and secreted isoforms.
  • CD166/ALCAM binds the T-cell surface receptor CD6, enhancing T-cell activation and proliferation. Related references: PMID:15048703, PMID:16352806, PMID:24945728
  • Homophilic ALCAM-ALCAM interactions contribute to immunological synapse maturation and dendritic cell–endothelial adhesion. Related references: PMID:15294938, PMID:15496415, PMID:16352806, PMID:23169771
  • ALCAM serves as a ligand for the inhibitory receptor LILRB4, potentiating LILRB4-mediated suppression of T-cell proliferation. Related references: PMID:29263213
  • ALCAM is required for the engraftment and maintenance of hematopoietic stem cells in the bone marrow. Related references: PMID:24740813
  • In endothelial cells, homotypic ALCAM interactions inhibit migration and promote tube formation, and the protein is essential for normal lymphatic vessel organization. Related references: PMID:15496415, PMID:23169771
  • A secreted splice variant competes with membrane-bound isoforms for interaction partners, inhibiting cell attachment and promoting endothelial migration.
  • CD166/ALCAM localizes to the cell membrane, cell projections (axons, dendrites), and is also secreted. It is expressed on activated T-cells, B-cells, monocytes, endothelial cells, and several tissues including spleen, placenta, and liver.

Experimental Guidance and Technical Tips

  • The antibody is supplied as an conjugate, ready for direct flow cytometry; no secondary detection reagent is needed.
  • Perform a titration (e.g., 0.1–5 µg per million cells) to determine the optimal staining concentration for your specific cellular system.
  • Human peripheral blood mononuclear cells (PBMCs) or CD166/ALCAM-expressing cell lines (such as activated T-cells) can serve as positive controls; include an appropriate isotype control.
  • Protect the antibody from light and store at 2–8°C; do not freeze.
  • For multicolor panels, verify fluorochrome compatibility with (excitation/emission: 488/520 nm).
  • The immunogen spans amino acids 28–526, covering the majority of the extracellular domain; consider validation for detection of both membrane-bound and soluble forms in your assay.
