Biological Background: CD163 Function and Localization

  • CD163, also known as M130 or hemoglobin scavenger receptor, is a member of the scavenger receptor cysteine-rich (SRCR) superfamily. It functions as an acute phase-regulated receptor on macrophages.
  • It mediates clearance and endocytosis of hemoglobin/haptoglobin complexes, protecting tissues from oxidative damage and facilitating iron recycling.
  • Binds hemoglobin/haptoglobin in a calcium- and pH-dependent manner, with higher affinity for multimeric haptoglobin phenotypes.
  • Upon ligand binding, CD163 triggers intracellular signals involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
  • The protein exists as membrane-bound and secreted (soluble sCD163) forms; sCD163 may have anti-inflammatory roles and serve as a biomarker for macrophage activation.
  • Subcellular localization includes the cell membrane and secreted. CD163 is predominantly expressed in tissue-resident macrophages (e.g., Kupffer cells, splenic red pulp macrophages) and monocytes, with isoform 3 being the major blood isoform.

  • For immunohistochemistry (IHC-P), antigen retrieval (e.g., citrate buffer, heat-induced) may be required; positive control tissues include liver, spleen, or inflamed tissues.
  • In immunofluorescence (IF-P), optimize permeabilization and blocking steps; CD163 localizes to the cell membrane and intracellular compartments.
  • For ELISA, the antibody may be suitable for capture or detection of soluble CD163; validate pair compatibility if used with other antibodies.
