Biological Background: CD162/PSGL-1 Function and Localization
- P-selectin glycoprotein ligand 1 (PSGL-1), also known as CD162, is encoded by the SELPLG gene.
- Functions as a SLe(x)-type proteoglycan that mediates leukocyte rolling on vascular surfaces through high-affinity, calcium-dependent interactions with E-, P-, and L-selectins, critical for the initial steps of inflammation.
- Acts as a receptor for enterovirus 71, facilitating viral entry.
- Localized to the plasma membrane as a type I transmembrane protein.
- Expressed on neutrophils, monocytes, and most lymphocytes.
- Undergoes post-translational modifications including sulfation, sialylation, phosphorylation, disulfide bond formation, and contains pyrrolidone carboxylic acid.
- Subject to alternative splicing and classified under host cell receptor for virus entry membrane proteins.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 42-295), enabling recognition of the native protein on cell surfaces.
- For Western blot, note that glycosylation may cause the apparent molecular weight to be higher than the predicted 43-44 kDa; consider deglycosylation for precise detection.
- Validate antibody reactivity in appropriate cell lines or tissues known to express PSGL-1, such as HL-60 or human leukocytes.
- For immunohistochemistry, optimize antigen retrieval and test a dilution range to minimize background.
- In immunofluorescence, a membrane staining pattern is expected; permeabilization may be omitted for surface detection.
- This antibody can be employed in ELISA for quantitative detection; pair with a compatible secondary antibody or use in a sandwich format.
