Biological Background: CD16 Function and Localization

  • CD16a (FCGR3A) is a low-affinity receptor for the Fc fragment of IgG, primarily expressed on natural killer (NK) cells and a subset of monocytes. It plays a central role in antibody-dependent cellular cytotoxicity (ADCC) and cytokine production. Related references: PMID:11711607, PMID:21768335, PMID:22023369, PMID:27670158
  • Upon binding to IgG-opsonized targets, CD16a triggers ITAM-mediated signaling cascades, leading to NK cell activation, degranulation, and target cell lysis. This process is critical for antiviral immunity and tumor surveillance. Related references: PMID:24412922, PMID:25786175, PMID:29967280, PMID:9916693, PMID:1825220, PMID:23024279, PMID:2532305
  • CD16a can also costimulate NK cells and mediate target cell lysis independently of IgG binding, highlighting its role in innate immune activation. Related references: PMID:10318937, PMID:23006327
  • The receptor mediates the antitumor efficacy of therapeutic monoclonal antibodies, with enhanced activity observed upon binding to afucosylated IgGs. Related references: PMID:27670158, PMID:34485821, PMID:28566370
  • In the context of Dengue virus infection, CD16a facilitates antibody-dependent enhancement (ADE) by binding to afucosylated IgG1-virus complexes, promoting viral entry into myeloid cells.
  • Subcellularly, CD16a is anchored at the cell membrane via a transmembrane helix and can also be found in a secreted form. It undergoes phosphorylation and glycosylation, which modulate its signaling functions.
  • Tissue expression is restricted to NK cells and a subset of monocytes, as confirmed at the protein level. Related references: PMID:2526846, PMID:27670158

Experimental Guidance and Technical Tips

  • The immunogen covers the extracellular domain (amino acids 20-208) of human CD16b, suggesting the antibody recognizes an epitope conserved between CD16a and CD16b. Consider validating cross-reactivity directly in your model system.
  • For Western blotting, a band near 29 kDa (non-glycosylated form) is expected, but higher molecular weight smears may appear due to heavy glycosylation. Using a reducing agent and optimizing gel percentage can improve resolution.
  • In IHC-P, anticipate membrane staining patterns; include appropriate negative and positive controls (e.g., NK cell-rich tissues such as spleen or tonsil).
  • For IF/ICC, fixation and permeabilization conditions should be optimized to preserve the extracellular epitope; live-cell staining may also be suitable for surface CD16.
  • ELISA applications may benefit from pairing with a detection antibody targeting a distinct epitope; consider the potential for afucosylated IgG binding interference in certain assay formats.
