Biological Background: CD155/PVR Function and Localization

  • CD155 (poliovirus receptor, PVR) is a member of the nectin family of immunoglobulin-like adhesion molecules, also known as nectin-like protein 5 (NECL-5). The PVR gene undergoes alternative splicing to generate multiple isoforms.
  • It functions as a critical NK cell adhesion molecule by binding CD96 and CD226. These interactions accumulate at the cell-cell contact site, promoting the formation of a mature immunological synapse that triggers NK cell cytotoxicity and IFN-gamma secretion.
  • CD155 plays a role in tumor cell invasion and migration. On tumor cells, high PVR expression can lead to fratricide NK cell activation, providing a mechanism of immune evasion.
  • As a host cell receptor for virus entry, CD155 mediates poliovirus infection by directing virion-PVR-containing endocytic vesicles to the microtubular network via interaction with DYNLT1, facilitating axonal retrograde transport. It also acts as a receptor for Pseudorabies virus.
  • Human cytomegalovirus (HHV-5) can prevent CD155 from reaching the cell surface, likely to escape immune recognition of infected cells by NK cells.
  • The 45 kDa transmembrane glycoprotein contains Ig-like domains and features disulfide bonds, phosphorylation sites, and a signal peptide. It localizes to the cell membrane and is also secreted, consistent with its roles in adhesion and signaling.

  • The immunogen corresponds to a region within amino acids 250–350 of CD155, which lies in the extracellular domain. This design may make the antibody suitable for detecting the extracellular portion in applications like ELISA.
  • For Western blot, the predicted molecular weight is approximately 45 kDa. Consider using a suitable gel percentage (e.g., 10% SDS-PAGE) and standard wet or semi-dry transfer conditions for proteins of this size.
  • As cross-reactivity is confirmed for human, validate the antibody in relevant human samples such as NK cells, tumor cell lines, or tissues expressing CD155.
