Biological Background: CD155 (PVR) Function and Localization
- CD155, also known as poliovirus receptor (PVR) or nectin-like protein 5 (NECL-5), is a member of the immunoglobulin superfamily involved in cell adhesion and immune recognition.
- The protein mediates natural killer (NK) cell adhesion and triggers effector functions by binding to two NK cell receptors, CD96 and CD226, leading to the formation of immunological synapses and the release of cytotoxic granules and IFN-gamma.
- CD155 plays a role in tumor cell invasion and migration, and its high expression on some tumors may enable immune evasion through fratricide NK cell activation or modular exchange of PVR molecules.
- As a crucial host cell receptor, CD155 facilitates the entry of poliovirus by interacting with the virion and targeting endocytic vesicles to the microtubular network via DYNLT1 for axonal retrograde transport. It also acts as a receptor for Pseudorabies virus.
- Human cytomegalovirus (HCMV) infection prevents CD155 from reaching the cell surface, likely as a mechanism to evade NK cell-mediated immune recognition.
