Biological Background: CD150/SLAM Function and Localization

  • CD150 (SLAMF1) is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, with a molecular weight of approximately 37 kDa. It is expressed as multiple isoforms, including a secreted form, due to alternative splicing.

  • As a self-ligand receptor, CD150 mediates homo- or heterotypic cell-cell interactions, modulating immune cell activation and differentiation. Its signaling is regulated by adaptors SH2D1A/SAP and SH2D1B/EAT-2, with distinct outcomes in T-cells and B-cells. In T-cells, CD150 signaling promotes proliferation and IFN-gamma production, while in B-cells, it activates ERK, AKT, and MAPK pathways. Related references: PMID:11806999 PMID:12458214 PMID:20231852

  • CD150 plays a role in innate immunity against Gram-negative bacteria by recognizing bacterial surface proteins OmpC/OmpF, regulating phagosome maturation and reactive oxygen species production. Related references: PMID:20818396

  • CD150 acts as a receptor for measles virus, facilitating viral entry into host cells.

  • CD150 is constitutively expressed on peripheral blood memory T-cells, immature thymocytes, and a proportion of B-cells, and is rapidly induced on naive T-cells upon activation. Related references: PMID:7617038

  • CD150 localizes primarily to the cell membrane, with a secreted isoform also present. It is a phosphoprotein and glycoprotein, with disulfide bonds important for its structure.

Experimental Guidance and Technical Tips

  • Since the immunogen is a recombinant fusion protein covering amino acids 21–237 of human CD150, this antibody targets the extracellular domain, making it suitable for flow cytometry applications detecting surface expression.
  • For flow cytometry, titrate the antibody to determine optimal staining concentration using relevant positive and negative control cells (e.g., activated T-cells or B-cell lines).
  • Include appropriate isotype controls and consider Fc receptor blocking to minimize non-specific binding.
  • Validate antibody performance in your specific experimental system, as antigen density and cell type may affect staining intensity.
