Biological Background: CD147/BSG Function and Localization

  • Basigin (BSG/CD147/EMMPRIN) is a highly glycosylated transmembrane protein belonging to the immunoglobulin superfamily. It functions as a receptor for extracellular cyclophilins, mediates monocarboxylate transporter localization, and plays critical roles in retinal development, tumor progression, and microbial infection. Related references: PMID:25957687, PMID:17127621, PMID:11688976
  • In the retina, BSG acts as a receptor for NXNL1, promoting cone photoreceptor survival by enhancing aerobic glycolysis via SLC16A1/GLUT1, and is essential for normal retinal maturation. Related references: PMID:25957687
  • BSG stimulates tumor growth, invasion, metastasis, and neoangiogenesis by inducing production of extracellular matrix metalloproteinases (MMPs) and upregulating VEGFA and KDR/VEGFR2 in both tumor and stromal cells. It also stimulates IL6 secretion in monocytes. Related references: PMID:21620857, PMID:11992541, PMID:12553375, PMID:15833850
  • As a signaling receptor for cyclophilins PPIA/CYPA and PPIB/CYPB, BSG regulates immune cell chemotaxis and adhesion. Related references: PMID:11688976, PMID:11943775
  • BSG functions as a coreceptor for VEGFR2 (KDR) in endothelial cells, enhancing VEGFA-mediated activation and downstream signaling, thereby promoting angiogenesis. Related references: PMID:25825981, PMID:19837976
  • BSG is an essential receptor for Plasmodium falciparum erythrocyte invasion, binding the parasite ligand RH5 and triggering intracellular calcium flux and cytoskeletal rearrangement. It also facilitates infection by SARS-CoV-1, HIV-1, measles virus, and human cytomegalovirus, but is not required for SARS-CoV-2 entry. Related references: PMID:22080952, PMID:26195724, PMID:28409866
  • Subcellularly, BSG localizes to the plasma membrane (including basolateral membrane in polarized cells), melanosomes, photoreceptor inner/outer segments, endosomes, and endoplasmic reticulum membrane. It is highly expressed in bone marrow, fetal liver, lung, testis, thymus, heart, kidney, skeletal muscle, and at the protein level in retinal cone photoreceptors, erythrocytes, and melanoma cell lines.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a sequence within the intracellular domain (aa 286-385) of human CD147/BSG (P35613). This region may be conserved, but cross-reactivity validation in other species is recommended before use.
  • For western blot (WB), a band around 42 kDa (glycosylated form) may be observed; consider using appropriate protein extraction and denaturing conditions to expose the epitope.
  • For immunohistochemistry (IHC-P), antigen retrieval and signal amplification steps may be necessary depending on tissue fixation conditions and target expression levels.
  • For ELISA, pair this antibody with a capture antibody targeting a distinct epitope to avoid competition; validate in the relevant sample matrix.
