Biological Background: CD146 Function and Localization
- CD146 (also known as MUC18 or gicerin) is a cell surface glycoprotein encoded by the Mcam gene, with alternative splicing generating multiple isoforms.
- Functions as a cell adhesion molecule, contributing to endothelial monolayer cohesion at intercellular junctions in vascular tissue.
- Its expression on melanoma cells may facilitate interactions with the vascular system, potentially enhancing metastatic spread.
- Participates in neural crest cell migration during embryonic development.
- Acts as a surface receptor, triggering tyrosine phosphorylation of FYN and PTK2/FAK and a transient increase in intracellular calcium (by similarity).
- Located at the plasma membrane as a single-pass type I membrane protein.
- Contains immunoglobulin-like domains; undergoes glycosylation, phosphorylation, and disulfide bond formation.
- Detected in melanoma cell lines.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 24–563, covering the majority of the extracellular domain; the antibody is expected to recognize the extracellular region of CD146.
- For Western blotting, a band at approximately 71 kDa (calculated from sequence) is expected; validate under reducing and non-reducing conditions as needed.
- Cross-reactivity has been confirmed with human and mouse samples; consider performing additional validation if testing other species.
- This polyclonal antibody may be suitable for ELISA; appropriate pairing with an anti-rabbit secondary antibody is recommended.
