Biological Background: CD146 Function and Localization
- Cell surface glycoprotein MUC18 (CD146), encoded by the MCAM gene, is also known as melanoma cell adhesion molecule, MUC18, HEMCAM, METCAM, MelCAM, and S-endo 1 endothelial-associated antigen.
- It functions as a cell adhesion molecule that plays a critical role in maintaining endothelial monolayer cohesion at intercellular junctions in vascular tissue.
- Its expression may facilitate melanoma cell interaction with vascular elements, promoting hematogeneous tumor spread and metastasis.
- Acts as a surface receptor triggering tyrosine phosphorylation of FYN and PTK2/FAK1, and a transient increase in intracellular calcium concentration.
- May be involved in neural crest cell migration during embryonic development.
- Predominantly localized to the membrane; tissue expression is detected in endothelial cells throughout the vasculature and in vascular smooth muscle of normal adult tissues.
- Strongly expressed in metastatic lesions and advanced primary melanomas, but rarely detected in benign melanocytic nevi and thin primary melanomas with low metastatic probability.
- Classified as an immunoglobulin domain-containing glycoprotein, with features including disulfide bonds, phosphorylation, and alternative splicing.
Experimental Guidance and Technical Tips
- As the antibody is raised against a peptide within the C-terminal region (a.a. 546–645), consider validating its reactivity in relevant sample systems.
- For Western blot, theoretical molecular weight is ~72 kDa; ensure appropriate gel percentage and transfer conditions.
- For ELISA, optimization of coating antigen and antibody dilution is recommended.
- Cross-reactivity with mouse and rat has been observed; verify species-specific detection in multi-species experiments.
