Biological Background: Cadherin-5 (CD144/VE-Cadherin) Function and Localization

  • Cadherin-5, also known as 7B4 antigen or vascular endothelial cadherin (VE-cadherin), is encoded by the CDH5 gene and belongs to the cadherin superfamily of calcium-dependent cell adhesion proteins.
  • Cadherins preferentially mediate homophilic interactions to connect adjacent cells, contributing to the sorting of heterogeneous cell types. Related references: PMID:21269602
  • In endothelial cells, cadherin-5 plays a critical role in controlling the cohesion and organization of intercellular junctions, thereby maintaining vascular integrity.
  • It associates with alpha-catenin, forming a physical link to the actin cytoskeleton. Related references: PMID:10861224
  • Cadherin-5 acts as a junctional anchor for AMOTL2 and MAGI1, coupling actin fibers to cell junctions in endothelial cells and regulating endothelial cell polarity and vascular lumen formation through the Par polarity complex and RAP1B. Related references: PMID:20332120
  • It positively regulates reorientation of actin stress fibers and endothelial cell reorientation in response to mechanical forces (mechanotransduction). Related references: PMID:25795300
  • Subcellular localization includes the cell junction (adherens junctions), cell membrane, and cytoplasm, with dynamic trafficking regulated by CTNND1/p120-catenin to control endocytosis.
  • Tissue expression is predominantly in endothelial cells (confirmed at protein level) and also in the brain. Related references: PMID:27338829, PMID:2059658
  • Characterized by alternative splicing, calcium-binding repeats, a transmembrane helix, and post-translational modifications including phosphorylation and glycosylation.

Experimental Guidance and Technical Tips

  • The immunogen comprises amino acids 48–593 of human Cadherin-5, which spans the extracellular domain. This design makes the antibody suitable for detecting cell surface CD144 by flow cytometry on live, non-permeabilized cells.
  • Because cadherin conformation is calcium-dependent, consider including physiological calcium concentrations (e.g., 1–2 mM) in staining and wash buffers to preserve native epitope structure.
  • For flow cytometry, validate the antibody using a known positive endothelial cell line such as HUVEC or EA.hy926, and use a CD144-negative cell line (e.g., HEK293) as a negative control.
  • Monoclonal antibodies provide high batch-to-batch consistency; however, each new lot should be titrated to determine the optimal staining concentration for your specific experimental system.
  • The conjugate is compatible with common violet laser lines (405 nm excitation) and can be multiplexed with other fluorophores; perform compensation controls as needed.
