Biological Background: CD144/VE Cadherin (Cadherin-5) Function and Localization
- Cadherin-5, also known as VE-cadherin or 7B4 antigen, is a calcium-dependent cell adhesion protein encoded by the CDH5 gene. It is a member of the cadherin superfamily and functions primarily as a homophilic adhesion receptor.
- It mediates calcium-dependent homophilic cell adhesion, contributing to the sorting of heterogeneous cell types. Related references: PMID:21269602
- Essential for endothelial cell cohesion and organization of intercellular junctions, it associates with alpha-catenin to anchor the actin cytoskeleton. Related references: PMID:10861224
- Together with KRIT1 and PALS1, it establishes and maintains correct endothelial cell polarity and vascular lumen by recruiting and activating the Par polarity complex and RAP1B. Related references: PMID:20332120
- Positively regulates reorientation of actin stress fibers and endothelial cell reorientation in response to cellular mechanotransduction. Related references: PMID:25795300
- Associates with CTNND1/p120-catenin to control CADH5 endocytosis, a mechanism that regulates cadherin surface levels.
- It localizes to cell junctions, specifically adherens junctions, as well as the cell membrane and cytoplasm.
- Expression is predominantly in endothelial cells (at the protein level) and also in the brain. Related references: PMID:27338829 ; PMID:2059658
- The protein is a glycoprotein and undergoes phosphorylation, as indicated by its keyword annotations, suggesting regulated signaling functions.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 48-593 of human CD144/VE Cadherin, spanning the extracellular domain, making this antibody well-suited for detecting surface epitopes on live cells by flow cytometry.
- As a directly conjugated antibody (), it can be used immediately without a secondary staining step. Prior to critical experiments, titrate the antibody to establish optimal concentrations for your specific cell types.
- Validate staining specificity using appropriate isotype controls and include positive (e.g., primary endothelial cells or HUVECs) and negative (e.g., CDH5-negative lines) controls.
- Consider fixation and permeabilization effects if intracellular staining is required, as the epitope is extracellular; test under both live and fixed conditions.
