Biological Background: CD14 Function and Localization

  • Monocyte differentiation antigen CD14 (CD14), also known as myeloid cell-specific leucine-rich glycoprotein, is a glycosylphosphatidylinositol (GPI)-anchored membrane glycoprotein belonging to the leucine-rich repeat (LRR) family.
  • CD14 acts as a coreceptor for bacterial lipopolysaccharide (LPS) in concert with LPS-binding protein (LBP), delivering LPS to the LY96/TLR4 complex to initiate innate immune responses (PubMed:16148141).
    Related references:
    PMID:16148141
  • Downstream signaling via MyD88, TIRAP, and TRAF6 leads to NF-kappa-B activation, cytokine secretion, and inflammatory response (PubMed:15895089, PubMed:8612135).
    Related references:
    PMID:15895089, PMID:8612135
  • CD14 serves as a coreceptor for TLR2:TLR6 and TLR2:TLR1 heterodimers in response to diacylated and triacylated lipopeptides, respectively; these complexes trigger signaling from the cell surface and traffic to the Golgi in a lipid-raft dependent manner.
  • As an accessory receptor, CD14 binds M.tuberculosis lipoproteins LprA, LprG, and LpqH in conjunction with TLR2 and TLR1, modulating antigen-presenting cell functions (PubMed:19362712).
    Related references:
    PMID:19362712
  • CD14 also binds electronegative LDL (LDL(-)) and mediates cytokine release induced by LDL(-).
  • Subcellular localization includes the cell membrane, membrane rafts, Golgi apparatus, and it can be secreted in a soluble form.
  • Detected on peritoneal macrophages, lung alveolar macrophages, dendritic macrophages, and lung macrophages at the protein level (PubMed:8612135, PubMed:19362712).

    PMID:8612135, PMID:19362712

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (residues 18-345) of mouse CD14, which encompasses the leucine-rich repeat region; this design can be useful for detecting both membrane-bound and soluble CD14 forms.
  • For Western blotting, the predicted molecular weight of CD14 is approximately 39 kDa; however, due to glycosylation, the observed band may run slightly higher. Consider using a suitable blocking and lysis buffer to preserve epitopes.
  • Flow cytometry can be applied to analyze CD14 cell surface expression on macrophages and monocytes; ensure proper gating and isotype controls for mouse samples.
  • ELISA may be suitable for detecting soluble CD14 or studying LPS-binding interactions; optimize coating conditions and detection antibodies for the specific assay format.
  • Validate the antibody in the relevant sample system, such as mouse peritoneal macrophages or lung macrophage populations, to confirm reactivity and specificity.
