Biological Background: CD14 Function and Localization
-
CD14 functions as a coreceptor for bacterial lipopolysaccharide (LPS), initiating the innate immune response to gram-negative bacteria.
Related references: PMID:1698311, PMID:23264655 -
In collaboration with LBP, CD14 binds monomeric LPS and transfers it to the LY96/TLR4 complex, triggering MyD88-dependent NF-kB activation and pro-inflammatory cytokine secretion.
Related references: PMID:20133493, PMID:22265692, PMID:23264655, PMID:8612135 -
CD14 also serves as a coreceptor for TLR2:TLR6 and TLR2:TLR1 heterodimers in response to diacylated and triacylated lipopeptides, respectively, facilitating signaling from the cell surface and subsequent trafficking to the Golgi.
Related references: PMID:16880211 -
Beyond LPS, CD14 binds electronegative LDL (LDL(-)) and mediates cytokine release induced by modified lipoproteins.
Related references: PMID:23880187 -
CD14 is localized to the cell membrane, membrane rafts, and Golgi apparatus; a soluble form is also secreted, expanding its functional range.
-
Tissue expression is predominantly on monocytes and macrophages, with weak detection on granulocytes.
Related references: PMID:1698311 -
The protein is heavily glycosylated and anchored to the membrane via a GPI linkage; it contains leucine-rich repeats and disulfide bonds critical for its structure and ligand recognition.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a sequence within amino acids 250-350 of human CD14; this region may be used for specific peptide blocking experiments to confirm antibody specificity.
- For immunofluorescence (IF/ICC and IF-P) and immunohistochemistry (IHC-P), optimize fixation and permeabilization protocols since CD14 is a membrane-anchored protein; antigen retrieval may be unnecessary for cell surface targets.
- In ELISA applications, consider using the antibody as a capture or detection reagent; validate its performance with recombinant CD14 or native samples from monocytic cell lines.
- Reported cross-reactivity is limited to human; if your experiments involve other species, confirm sequence homology of the epitope region and test cross-reactivity empirically.
