Biological Background: CD13/ANPEP Function and Localization

  • Aminopeptidase N, also designated APN, CD13, alanyl aminopeptidase, or gp150, is a zinc-dependent type II transmembrane metalloprotease encoded by the ANPEP gene.
  • Functions as a broad-specificity aminopeptidase in the final digestion of peptides derived from gastric and pancreatic proteolysis; processes peptide hormones including angiotensin III and IV, neuropeptides, and chemokines.
  • May cleave peptides presented on MHC class II molecules of antigen-presenting cells, contribute to angiogenesis, promote cholesterol crystallization, and regulate amino acid transport by interacting with the SLC6A19 transporter.
  • Localizes to the cell membrane and is expressed in epithelial cells of kidney, intestine, and respiratory tract, as well as on granulocytes, monocytes, endothelial cells, and certain CNS synaptic membranes; a soluble form is detectable in plasma, often elevated in cancer patients.
  • Serves as a cellular receptor for human coronavirus 229E (HCoV-229E) spike glycoprotein and mediates entry of human cytomegalovirus (HCMV).
  • Annotated with keywords including aminopeptidase, angiogenesis, cell membrane, host cell receptor for virus entry, metalloprotease, protease, transmembrane, and zinc.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular region (aa 69-967) of human CD13, supporting detection of surface‑exposed epitopes in flow cytometry and immunohistochemistry.
  • For western blot, anticipate a band around 110 kDa; validate specificity using a positive control such as kidney or intestinal tissue lysate and an appropriate molecular weight marker.
  • In ELISA applications, consider pairing with a second antibody that recognizes a non‑overlapping epitope to develop a robust sandwich assay.
  • Optimize antigen retrieval protocols for IHC‑P, as CD13 is a transmembrane protein, and validate staining patterns in relevant normal and tumor tissues.
