Biological Background: CD133 Function and Localization
- CD133, also known as Prominin-1, is encoded by the Prom1 gene in mouse (also designated Proml1). Alternative splicing produces multiple isoforms.
- Plays a role in cell differentiation, proliferation and apoptosis; binds cholesterol in plasma membrane microdomains and contributes to apical membrane organization.
- Functions as a key regulator of disk morphogenesis during early retinal development. Related references: PMID:19228982
- Involved in regulation of MAPK and Akt signaling pathways; in neuroblastoma cells, suppresses cell differentiation such as neurite outgrowth in a RET-dependent manner.
- Localizes to the apical cell membrane, microvillus membrane, cilium, photoreceptor outer segment, endoplasmic reticulum, and ER-Golgi intermediate compartment.
- Exhibits tissue-specific expression: in adult, expressed in kidney tubules and brain ependymal layer, but not in gut, liver, lung, pituitary, adrenal, heart or spleen; in embryo, present on apical side of neuroepithelial cells, lung buds, gut, ureter buds.

- The immunogen used is a recombinant fusion protein covering amino acids 20-107 of mouse CD133. For WB, expect a band at approximately 97 kDa. Consider using tissue or cell lysates where CD133 is known to be expressed, such as kidney or retina.
- For immunofluorescence (IF-P), targeting apical membranes or primary cilia in epithelial cells may be relevant. Validate staining patterns in the appropriate cellular context.
- In immunoprecipitation (IP) assays, ensure the antibody is compatible with the chosen lysis buffer and detection system. Pre-clearing and proper controls are recommended.
