Biological Background: CD133 Function and Localization

  • CD133, also known as Prominin-1, is encoded by the Prom1 gene in mouse (also designated Proml1). Alternative splicing produces multiple isoforms.
  • Plays a role in cell differentiation, proliferation and apoptosis; binds cholesterol in plasma membrane microdomains and contributes to apical membrane organization.
  • Functions as a key regulator of disk morphogenesis during early retinal development. Related references: PMID:19228982
  • Involved in regulation of MAPK and Akt signaling pathways; in neuroblastoma cells, suppresses cell differentiation such as neurite outgrowth in a RET-dependent manner.
  • Localizes to the apical cell membrane, microvillus membrane, cilium, photoreceptor outer segment, endoplasmic reticulum, and ER-Golgi intermediate compartment.
  • Exhibits tissue-specific expression: in adult, expressed in kidney tubules and brain ependymal layer, but not in gut, liver, lung, pituitary, adrenal, heart or spleen; in embryo, present on apical side of neuroepithelial cells, lung buds, gut, ureter buds.
