  • CD13 (cluster of differentiation 13), also designated Aminopeptidase N (AP-N), is a type II integral membrane zinc-dependent metalloprotease widely known as an alanyl aminopeptidase. Alternate names include Aminopeptidase M and membrane protein p161.
  • As a broad-specificity aminopeptidase, CD13 catalyzes the removal of N-terminal amino acids from peptides, contributing to the final stages of protein digestion and the processing of regulatory peptides such as angiotensin III/IV, neuropeptides, and chemokines (By similarity).

  • The protein is subject to N-linked glycosylation, tyrosine sulfation, and phosphorylation; it also contains disulfide bonds and a zinc ion coordinated in the active site, consistent with its metalloprotease function.

  • For IHC-P, optimal antigen retrieval should be determined empirically; common protocols include heat-induced epitope retrieval in citrate (pH 6) or EDTA (pH 9) buffers.
