Biological Background: CD13 Function and Localization
- CD13 (cluster of differentiation 13), also designated Aminopeptidase N (AP-N), is a type II integral membrane zinc-dependent metalloprotease widely known as an alanyl aminopeptidase. Alternate names include Aminopeptidase M and membrane protein p161.
- As a broad-specificity aminopeptidase, CD13 catalyzes the removal of N-terminal amino acids from peptides, contributing to the final stages of protein digestion and the processing of regulatory peptides such as angiotensin III/IV, neuropeptides, and chemokines (By similarity).
- In antigen-presenting cells, CD13 is implicated in the trimming of peptides bound to major histocompatibility complex class II molecules, a process that may influence adaptive immune responses (PubMed:8691132). Related references: PMID:8691132
- CD13 has been reported to play a role in amino acid homeostasis by interacting with the amino acid transporter SLC6A19 and modulating its activity at the plasma membrane (PubMed:22677001). Related references: PMID:22677001
- Localized predominantly at the cell surface, CD13 is expressed in a variety of tissues including the intestinal brush border, kidney proximal tubules, liver, lymph node, spleen, brain, and on cells of the monocyte-macrophage lineage, dendritic cells, and B lymphocytes, but not on T cells.
- The protein is subject to N-linked glycosylation, tyrosine sulfation, and phosphorylation; it also contains disulfide bonds and a zinc ion coordinated in the active site, consistent with its metalloprotease function.

- As the immunogen covers a large extracellular fragment (amino acids 69–966), the resulting polyclonal antibody likely recognizes multiple epitopes and can be used to detect both native and denatured CD13.
- In Western blotting, CD13 migrates as a broad band typically above the predicted 109 kDa due to N-linked glycosylation; optimization of gel percentage and transfer conditions is recommended.
- For IHC-P, optimal antigen retrieval should be determined empirically; common protocols include heat-induced epitope retrieval in citrate (pH 6) or EDTA (pH 9) buffers.
- The antibody is compatible with ELISA applications; as a polyclonal reagent, it may serve as a capture or detection antibody in immunoassay development.
- Given the reported cross-reactivity with human and rat CD13, validation in the target species and sample matrix is essential for assay specificity.
