Biological Background: CD11b Function and Localization
- CD11b (integrin alpha-M), encoded by the ITGAM gene, is the alpha chain of the MAC-1 integrin complex, also known as CR-3, MO1, and CD11B (PubMed:1346576).
- ITGAM/ITGB2 integrin mediates diverse adhesive interactions in monocytes, macrophages, and granulocytes, including uptake of complement-coated particles via recognition of the iC3b fragment. Related references: PMID:20008295, PMID:9558116
- This integrin serves as a receptor for fibrinogen (recognizing P1 and P2 peptides of the gamma chain) and factor X, facilitating immune cell adhesion and migration.
- It regulates neutrophil migration and, in association with ITGB2, is required for CD177-PRTN3-mediated activation of TNF-primed neutrophils. Related references: PMID:28807980, PMID:21193407
- As a receptor for ICAM1, it mediates adhesion between stimulated neutrophils and endothelial cells, a key step in inflammation. Related references: PMID:1980124
- Subcellular localization is primarily at the cell membrane and membrane rafts, consistent with its role in signal transduction.
- Predominantly expressed in monocytes and granulocytes, including neutrophils, supporting its central function in innate immunity.
- The protein undergoes alternative splicing and post-translational modifications such as glycosylation and disulfide bond formation, which are critical for its structural integrity and function.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal sequence (aa 1-100) of human CD11b, which may confer high specificity for the full-length protein.
- For Western Blot, a band at approximately 127 kDa is expected; consider using reducing conditions and validate with positive control lysates from human monocytes or neutrophils.
- In immunohistochemistry (IHC-P), antigen retrieval and blocking steps may be optimized to reduce background in formalin-fixed paraffin-embedded tissues.
- For ELISA, the antibody can be used as a capture or detection reagent; ensure proper pairing validation for sandwich immunoassays.
