Biological Background: CD11b Function and Localization
- Integrin alpha-M (CD11b), encoded by the Itgam gene, is a member of the integrin alpha chain family and combines with the beta-2 subunit (ITGB2/CD18) to form the complement receptor CR3 (Mac-1). Alternative splicing yields multiple isoforms.
- The ITGAM/ITGB2 integrin mediates adhesive interactions of monocytes, macrophages, and granulocytes, including binding to the iC3b fragment of complement C3 and facilitating uptake of complement-coated particles and pathogens (By similarity).
- Beyond complement, CD11b serves as a receptor for fibrinogen, factor X, and ICAM1; it recognizes RGD motifs and the fibrinogen gamma chain P1/P2 peptides, thereby promoting leukocyte adhesion and migration.
- The receptor plays a central role in neutrophil migration and, together with CD18, is required for CD177-PRTN3-mediated activation of TNF-primed neutrophils (By similarity).
- It may regulate phagocytosis-induced apoptosis in extravasated neutrophils, contributing to the resolution of inflammation (By similarity).
- In the brain, CD11b is required together with TYROBP/DAP12 in microglia for production of superoxide ions that drive developmental neuronal apoptosis. Related references: PMID:18685038
- Subcellularly, CD11b is localized at the cell membrane and is enriched in membrane rafts.
- Tissue-specific expression is predominantly observed in monocytes, granulocytes, a subset of peritoneal mast cells, and microglia.

- The immunogen used to generate this antibody corresponds to amino acids 143–337 of mouse CD11b, covering a portion of the extracellular N-terminal region. This design may allow recognition of both native and denatured protein conformations, making it suitable for Western blot, immunohistochemistry, and ELISA.
- For Western blot analysis, CD11b migrates at approximately 127 kDa. Consider using a 6–12% SDS-PAGE gel and appropriate positive controls (e.g., macrophage or granulocyte lysates) to verify specificity.
- Due to the predicted glycosylation and disulfide-bonded structure, reducing conditions and optimized sample preparation may influence detection. Validate the antibody in the relevant sample system to confirm performance.
- This antibody is reported to cross-react with human and rat CD11b; however, sequence differences may affect affinity in these species, so independent validation in the target species is recommended.
- For IHC-P applications, antigen retrieval protocols and tissue fixation methods should be optimized for the specific sample type to achieve the best signal-to-noise ratio.
- In ELISA, the antibody can be used for capture or detection of CD11b; consider pairing it with a compatible monoclonal antibody in a sandwich ELISA format to enhance sensitivity.
