Biological Background: Integrin alpha-L (ITGAL/CD11a) Function and Localization
- Integrin alpha-L, encoded by ITGAL (CD11A), is the alpha subunit of the leukocyte-specific integrin lymphocyte function-associated antigen-1 (LFA-1), which pairs with integrin beta-2 (CD18). It belongs to the integrin alpha chain family and is characterized by multiple domain repeats and calcium/magnesium-binding motifs.
- LFA-1 serves as a receptor for intercellular adhesion molecules ICAM1, ICAM2, ICAM3, and ICAM4, mediating leukocyte-endothelial interactions and T-cell adhesion. Related references: PMID:10846180, PMID:12526797, PMID:1676048
- It also binds F11R and the ubiquitin-like protein ISG15, linking to tight junction regulation and antiviral responses. Related references: PMID:11812992, PMID:15528364, PMID:29100055
- LFA-1 is critical for immune synapse formation, translating TCR engagement and ICAM1 density into graded adhesion, and contributes to cytotoxic T-cell and natural killer cell killing. Related references: PMID:38195629, PMID:15356110
- Through ICAM3 binding, ITGAL participates in the phagocytosis of apoptotic neutrophils by macrophages. Related references: PMID:23775590
- Integrin alpha-L localizes to the cell membrane and membrane rafts, and is primarily expressed on leukocytes, including T-cells, neutrophils, and monocytes.
- It is involved in lymphopoiesis, as required for generation of common lymphoid progenitor cells in bone marrow.
- Post-translational modifications include phosphorylation, glycosylation, and disulfide bonds; the protein is also subject to alternative splicing.
Experimental Guidance and Technical Tips
- Because the immunogen is a synthetic peptide within amino acids 1071-1170, the antibody may recognize the C-terminal region. Consider epitope accessibility in native versus denaturing conditions.
- For Western blotting, a band around 129 kDa is expected; note that glycosylation may cause slight shifts.
- Validate reactivity in the relevant sample system (human leukocyte cell lines or primary cells) and include appropriate controls.
- For IHC-P, antigen retrieval may be necessary; optimize using human lymphoid tissue sections.
- ELISA applications may require pairing with a detection antibody; titrate to ensure specificity.
