Biological Background: Integrin alpha-L Function and Localization
- Integrin alpha-L (ITGAL), also known as CD11a or LFA-1 alpha chain, is a leukocyte-specific transmembrane protein that associates with ITGB2 (CD18) to form the lymphocyte function-associated antigen 1 (LFA-1) integrin.
- LFA-1 localizes to the plasma membrane and lipid rafts on immune cells.

- The immunogen (aa 26-340 of human CD11a) targets the N-terminal extracellular I-domain, which is involved in ligand binding; this may result in antibodies that recognize native epitopes. Verify reactivity in the relevant sample system.
- For Western blotting, the CD11a protein runs at approximately 129 kDa (uniprot theoretical), with additional bands at 118 and 134 kDa likely reflecting glycosylation and alternative isoforms. Optimize lysis, loading, and transfer conditions for high-molecular-weight proteins.
- In flow cytometry, use freshly isolated leukocytes and include Fc receptor blocking and isotype controls. Surface staining of live cells is recommended to preserve epitope integrity.
- The antibody is validated on human samples only; cross-reactivity with other species should be empirically tested.
