Biological Background: CD119 Function and Localization

  • CD119, also known as Interferon gamma receptor 1 (IFNGR1) or IFN-gamma-R-alpha, is a single-pass type I membrane protein that serves as the ligand-binding subunit of the interferon gamma receptor.
  • The receptor plays a crucial role in mediating antimicrobial, antiviral, and antitumor immune responses by activating effector immune cells and enhancing antigen presentation. Related references: PMID:20926559, PMID:27286456
  • CD119 associates with the accessory factor IFNGR2 to form a functional receptor complex capable of signal transduction. Related references: PMID:2137461, PMID:2530216, PMID:2530582
  • Upon interferon gamma binding, the intracellular domain of CD119 undergoes a conformational change that permits the recruitment and activation of JAK1 and JAK2 kinases; activated JAK1 then phosphorylates CD119, creating a docking site for STAT1. Related references: PMID:19889125
  • Phosphorylated STAT1 dimerizes and translocates to the nucleus to stimulate transcription of interferon-gamma-responsive genes. STAT3 can also be activated via this pathway, although to a lesser extent. Related references: PMID:15284232
  • The phosphorylated intracellular domain also provides a binding site for SOCS1, which negatively regulates JAK-STAT signaling by competing with STAT1 for receptor docking. Related references: PMID:15522878
  • CD119 is localized primarily at the cell membrane and contains structural features including an immunoglobulin-like domain, disulfide bonds, glycosylation sites, and a transmembrane helix; it is also subject to phosphorylation and ubiquitin conjugation.
  • The mature protein has a calculated molecular weight of approximately 52.3 kDa, consistent with the observed band in Western blotting.

Experimental Guidance and Technical Tips

  • For Western blotting, use appropriate protein extraction and reducing conditions; the expected band is ~52 kDa. Perform a dilution series to determine the optimal primary antibody concentration for your specific sample type.
  • For ELISA applications, consider using the antibody as a capture or detection reagent; a checkerboard titration with recombinant mouse CD119 or cell lysates is recommended to establish working concentrations.
  • This antibody has been validated for use with mouse samples; cross-reactivity with other species has not been tested and should be verified experimentally if needed.
  • As a polyclonal antibody, lot-to-lot consistency should be confirmed in pilot experiments when switching to a new batch.
