Biological Background: CD117/c-Kit Function and Localization
- KIT encodes the mast/stem cell growth factor receptor Kit, a transmembrane tyrosine-protein kinase that acts as a cell-surface receptor for the cytokine KITLG/SCF. It is also known as proto-oncogene c-Kit, p145 c-kit, and CD117.
- Upon ligand binding, KIT activates multiple signaling pathways including PI3K/AKT1, RAS/RAF/MAPK, and phospholipase C gamma (PLCG1), leading to the production of second messengers DAG and IP3. It phosphorylates substrates like PIK3R1, PLCG1, CBL, SRC, and SHC1, and promotes STAT transcription factor activation.
- KIT is essential for a wide range of biological processes: cell survival and proliferation, hematopoiesis, stem cell maintenance, gametogenesis, melanogenesis, and mast cell development, migration, and function.
- Subcellularly, the receptor is located primarily in the cell membrane but can also be found in the cytoplasm. Tissue-specific expression includes spermatogonia and Leydig cells in the testis, and it is maintained in ejaculated spermatozoa. Related references: PMID:20601678
- The protein undergoes post-translational modifications such as glycosylation, disulfide bond formation, and phosphorylation (tyrosine kinase activity), and is subject to ubiquitination, internalization, and degradation upon activation.
- Alternative splicing yields multiple isoforms; disease variants are associated with piebaldism and gastrointestinal stromal tumors. It belongs to the immunoglobulin domain-containing receptor tyrosine kinase family.

- This antibody, raised against the extracellular domain (aa 26-520), is suitable for Western blotting (WB) and ELISA. In WB, a band at approximately 110 kDa is expected under reducing conditions.
- Cross-reactivity is confirmed for human samples; the suitability for other species should be experimentally determined.
