Biological Background: CD117/c-Kit Function and Localization
- Protein identity: CD117/c-Kit is the mast/stem cell growth factor receptor Kit, a receptor tyrosine kinase encoded by the Kit gene (mouse GeneID: 16590). It is also known as proto-oncogene c-Kit.
- Core function: Acts as the cell-surface receptor for the cytokine KITLG/SCF and plays essential roles in cell survival, proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration and function, and melanogenesis.
- Signaling pathways: Upon binding SCF, KIT activates multiple signaling cascades including PI3K/AKT, RAS/MAPK, PLCγ-mediated calcium mobilization, and JAK/STAT pathways. It phosphorylates key adaptors and enzymes such as PIK3R1, PLCG1, CBL, and SRC.
- Isoforms and tissue specificity: Alternative splicing generates several isoforms. Isoform 1 is widely expressed (fetal liver, bone marrow). Isoform 3 is enriched in hematopoietic stem cells. Protein-level detection shows isoform 1 and 2 in bone marrow, spermatogonia, and spermatocytes; isoform 3 in round/elongating spermatids and spermatozoa.
- Subcellular localization: Primarily located at the cell membrane; also found in the cytoplasm.
- Post-translational modifications and key structural features: The receptor is a glycoprotein with disulfide bonds, immunoglobulin domains, and a cytoplasmic tyrosine kinase domain. It undergoes phosphorylation and ubiquitination, which regulate signaling and degradation.
- Keywords: Proto-oncogene, tyrosine-protein kinase, ATP-binding, magnesium/metal-binding, transmembrane helix, Ubl conjugation, repeat, signal.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein corresponding to mouse CD117/c-Kit. For Western blot, the predicted molecular weight of the full-length protein is ~109 kDa; a band around this size is typically observed. The provided protein weight also notes 23 kDa, possibly representing the recombinant fragment.
- When performing WB, consider using reducing conditions and appropriate positive controls (e.g., mouse mast cell or hematopoietic cell lysates). Endogenous expression in tissues should be verified in the relevant sample system.
- For ELISA, the recombinant immunogen can be used as a coating antigen or standard, but optimal coating conditions and detection ranges must be determined empirically.
- Validate the antibody’s performance in your specific application and sample matrix, as conditions may affect specificity and sensitivity.
