Biological Background: CD117/c-Kit Function and Localization

• CD117/c-Kit, also known as mast/stem cell growth factor receptor Kit, is a receptor tyrosine kinase encoded by the KIT proto-oncogene. It is a single-pass transmembrane glycoprotein of the immunoglobulin superfamily.
• Upon binding its ligand, stem cell factor (KITLG/SCF), Kit dimerizes and activates multiple downstream signaling cascades, including PI3K-AKT, RAS-MAPK, and JAK-STAT pathways, regulating cell survival, proliferation, and differentiation.
• Kit plays critical roles in hematopoiesis, stem cell maintenance, gametogenesis, melanogenesis, and mast cell development and function.

• For Western blotting (WB), the predicted molecular weight of unglycosylated Kit is ~110 kDa, but the mature glycoprotein often migrates at higher molecular weights (~145 kDa) under reducing conditions. Validate in your sample.
• This polyclonal antibody recognizes human, mouse, and rat Kit. Consider cross-reactivity testing for other species if needed.
• Kit is expressed in membrane and cytoplasm; thus, permeabilization is necessary for immunofluorescence or immunohistochemistry to detect intracellular pools.
