Biological Background: CD117/c-Kit Function and Localization

• Mast/stem cell growth factor receptor Kit (also known as SCFR, c-Kit, CD117, PBT) is a tyrosine-protein kinase encoded by the KIT gene, a proto-oncogene with multiple isoforms.
• Functions as the cell-surface receptor for stem cell factor (KITLG/SCF) and regulates cell survival, proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration, and melanogenesis.
• Upon ligand binding, KIT autophosphorylates and activates downstream signaling cascades including PI3K/AKT1, RAS/RAF/MAPK, PLCγ/PKC, and JAK/STAT pathways.
• Localizes to the cell membrane but can also be found in the cytoplasm, and undergoes ligand-induced internalization and degradation.

• For Western blotting, expect a band at approximately 110 kDa. Use a reducing SDS-PAGE sample buffer and validate detection with a known CD117-positive cell line (e.g., HMC-1, K562).
• In immunofluorescence (IF/ICC) and immunohistochemistry (IHC‑P), antigen retrieval (e.g., heat‑induced epitope retrieval in citrate buffer pH 6.0) and blockade of endogenous peroxidase activity may be required. Optimize primary antibody concentration and incubation conditions.
• For ELISA applications, the antibody may be used as a capture or detection antibody. Pairing with a validated ligand or antibody set is advised; confirm specificity with recombinant CD117 protein.
