Biological Background: CD115/CSF-1R Function and Localization
- CD115/CSF-1R encodes the macrophage colony-stimulating factor 1 receptor, a cell-surface tyrosine-protein kinase also known as CSF-1R, M-CSF-R, and proto-oncogene c-Fms (alternative aliases: FMS, CSFR, FIM2, GPSC, HDLS, C-FMS, CD115, HDLS1, BANDDOS).
- Serves as the receptor for both CSF1 and IL34, critically regulating the survival, proliferation, and differentiation of mononuclear phagocytes such as macrophages and monocytes.
- Plays a key role in innate immunity and inflammatory responses by promoting the release of pro-inflammatory chemokines upon ligand engagement.
- Essential for osteoclast proliferation and differentiation, bone resorption, and normal bone/tooth development; also required for male/female fertility and mammary gland morphogenesis during pregnancy.
- Ligand binding triggers multiple signaling cascades—ERK1/2, JNK, AKT1, STAT3/5, SRC kinases—and results in phosphorylation of PIK3R1, PLCG2, GRB2, SLA2, CBL; signaling is down-regulated by phosphatases and receptor internalization. Related references: PMID:20504948, PMID:30982609
- In the central nervous system, may contribute to the development of microglia macrophages. Related references: PMID:30982608
- Localizes to the cell membrane and is predominantly expressed in bone marrow and differentiated blood mononuclear cells; post-translational modifications include glycosylation, disulfide bonds, and ubiquitination, with features typical of immunoglobulin-domain-containing receptors.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning the C‑terminal region (aa 873‑972) of human CD115/CSF-1R; this region is likely conserved across variants, but sequence alignment should be performed to predict reactivity with specific isoforms.
- For Western blot, a major band near 108 kDa is anticipated; under reducing and denaturing conditions, confirm the band in positive controls such as macrophage cell lysates (e.g., THP‑1, RAW 264.7) and validate with appropriate blocking and antibody dilution.
- In immunofluorescence and flow cytometry, expect cell‑surface staining; fixation and permeabilization protocols should preserve the epitope. Co‑staining with a membrane marker is recommended to confirm localization.
- The product is cited as cross‑reacting with human, mouse, and rat; however, lot‑to‑lot variability is possible with polyclonal antibodies—re‑validate in your own experimental system and species of interest before proceeding to large‑scale or quantitative assays.
- For ELISA applications, optimize the coating antigen and antibody concentration; the polyclonal nature may improve sensitivity but may also increase background if not titrated properly.
