Biological Background: CD112 (Nectin-2) Function and Localization
- CD112, also known as Nectin-2, herpes virus entry mediator B (HveB), and poliovirus receptor-related protein 2, is a cell adhesion molecule belonging to the nectin family.
- It functions as a modulator of T-cell signaling, acting either as a costimulator or coinhibitor depending on the receptor it binds. Interaction with CD226 stimulates T-cell proliferation and production of cytokines (IL2, IL5, IL10, IL13, IFNG), while interaction with PVRIG inhibits T-cell proliferation.
- CD112 is a probable cell adhesion protein, playing a role in cell-cell interactions.
- The protein localizes to the cell membrane, where it functions as a transmembrane receptor.
- Tissue specificity: expressed in brain, spinal cord, spleen, kidney, heart, and liver.
- Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation; it contains immunoglobulin-like domains characteristic of the nectin family.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 32-345) of mouse CD112, which is well conserved between mouse and human (validated cross-reactivity). This region includes the immunoglobulin-like domains critical for ligand binding.
- For Western blot (WB), consider reducing conditions and validate in relevant mouse tissue lysates (e.g., spleen, brain) or human cell lines; the predicted molecular weight is ~57 kDa (UniProt: 57318 Da).
- In ELISA applications, the polyclonal nature allows flexibility for use as capture or detection reagent; we recommend titrating the antibody individually for each assay system.
- Given the dual role in T-cell signaling, this antibody may be useful in functional studies investigating immune checkpoint modulation (CD226 vs. PVRIG pathways).
