Biological Background: Neprilysin Function and Localization
- Neprilysin (CD10/MME) is a type II integral membrane protein belonging to the M13 family of zinc metallopeptidases. It is also known as common acute lymphocytic leukemia antigen (CALLA), enkephalinase, and neutral endopeptidase (NEP).
- It exhibits thermolysin-like specificity, primarily acting on polypeptides of up to 30 amino acids by cleaving peptide bonds on the amino side of hydrophobic residues. Related references: PMID:15283675, PMID:6208535, PMID:6349683
- Neprilysin plays a critical role in the inactivation of opioid peptides Met- and Leu-enkephalins by hydrolyzing the Gly-Phe bond. Related references: PMID:17101991, PMID:6349683

Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 52–750), making this antibody suitable for detecting native neprilysin on cell surfaces.
- For Western blotting, a band around 86 kDa is expected. Consider using reducing conditions and appropriate loading controls.
- For immunohistochemistry on paraffin-embedded tissues (IHC-P), antigen retrieval steps may enhance detection. Validate staining patterns with known positive and negative controls.
- In ELISA applications, this antibody can serve as a capture or detection reagent when paired with a second CD10/MME antibody recognizing a non-overlapping epitope.
