Biological Background: CD107b/LAMP2 Function and Localization
- LAMP2 is a lysosomal membrane glycoprotein essential for lysosome biogenesis, pH regulation, and autophagy (PMID:10972293; PMID:30718432). Related references: PMID:10972293, PMID:30718432
- It mediates chaperone-mediated autophagy (CMA) by binding target proteins such as GAPDH, GPX4, NLRP3, and MLLT11, targeting them for lysosomal degradation (PMID:30718432). Related references: PMID:30718432
- Required for autophagosome-lysosome fusion, LAMP2 facilitates the accumulation of STX17 on autophagosomes (PMID:27628032). Related references: PMID:27628032

Experimental Guidance and Technical Tips
- The immunogen covers amino acids 26–189 of mouse LAMP2, corresponding to the N-terminal luminal domain with multiple N-glycosylation sites. This region may present both linear and conformational epitopes, suitable for detection in denaturing (WB) and native (ELISA) conditions.
- For Western blot, the predicted molecular weight is ~46 kDa, but extensive glycosylation can cause smearing or a larger apparent size. Validate specificity using mouse tissue lysates such as liver or kidney.
- This polyclonal antibody may recognize various LAMP2 splice isoforms (Lamp-2a, Lamp-2b, Lamp-2c). Confirm isoform reactivity in the relevant biological context.
- ELISA applications should be optimized by titrating the antibody and appropriate blocking; cross-reactivity remains limited to mouse according to the supplier's specifications.
