Biological Background: CD102 (ICAM2) Function and Localization
- CD102, also known as intercellular adhesion molecule 2 (ICAM2), is a member of the immunoglobulin superfamily and a cell adhesion molecule encoded by the ICAM2 gene.
- Functions as a receptor ligand for the integrin LFA-1 (ITGAL:ITGB2) to mediate leukocyte cell-cell adhesion on resting cells. Related references: PMID:10077629 PMID:1676048 PMID:2497351
- Also serves as an endothelial ligand for dendritic cell receptor CD209, triggering transendothelial migration of DC precursors from blood into peripheral tissues and subsequently into lymphoid organs. Related references: PMID:11017109
- Mediates adhesive interactions important for antigen presentation, lymphocyte recirculation, and immune surveillance.
- Constitutively expressed on leukocytes (resting lymphocytes, monocytes, but not neutrophils) and endothelial cells, with abundant expression on vascular endothelial cells and lymphatic vessels in tonsils. Related references: PMID:1676048 PMID:11017109
- Localizes to the cell membrane and microvilli, consistent with its role in mediating cell-cell contact and migration.
- Contains immunoglobulin-like domains, disulfide bonds, and is N-glycosylated, with a predicted molecular weight of approximately 30.7 kDa.
Experimental Guidance and Technical Tips
- Immunogen design: The antibody was raised against recombinant human CD102 protein; epitope may include extracellular Ig domains critical for ligand binding.
- For Western blot (WB): The predicted molecular weight of ICAM2 is ~30 kDa. Consider optimizing reducing conditions to prevent aggregation due to disulfide bonds.
- For IHC-P: Antigen retrieval may be required; validate staining in positive control tissues known to express CD102, such as tonsil or vascular endothelium.
- For ELISA: Consider pairing with a detection antibody that recognizes a non-overlapping epitope to avoid competitive interference.
- Cross-reactivity with mouse and rat ICAM2 suggests potential utility in animal models; however, verify sequence homology and binding affinity in the specific experimental system.
