Biological Background: CD101/IGSF2 Function and Localization
- CD101, also known as immunoglobulin superfamily member 2 (IGSF2), cell surface glycoprotein V7, and EWI-101, is a transmembrane protein encoded by the CD101 gene in humans.
- It functions as an inhibitor of T-cell proliferation induced by CD3, suppressing IL2RA expression and IL2 secretion in activated T-cells.
- CD101 inhibits tyrosine kinases required for IL2 production and cellular proliferation, and blocks phospholipase C-gamma-1 (PLCG1) phosphorylation, preventing CD3-induced calcium flux.
- It prevents nuclear translocation of the nuclear factor of activated T-cells, further contributing to its immunosuppressive role.
- CD101 plays a role in T-cell inhibition through IL10 secretion by cutaneous dendritic cells, and is a potential marker of CD4+ CD56+ leukemic tumor cells.
- Subcellularly, CD101 localizes to the plasma membrane as a single-pass type I membrane protein.
- Tissue expression is detected in lung, thymus, small intestine, cutaneous dendritic cells, activated T-cells, monocytes, granulocytes, and certain leukemic cells, including Langerhans cells from Langerhans cell histiocytosis patients.
- It belongs to the immunoglobulin superfamily and contains multiple Ig-like domains, with post-translational modifications including disulfide bonds and glycosylation.

- The immunogen covers amino acids 21–410, corresponding to the extracellular region, making this antibody suitable for detecting membrane-bound CD101 in live-cell FC or IF/ICC, as well as soluble forms in ELISA.
- For WB, the predicted molecular weight is ~115 kDa; consider using a 6–8% SDS-PAGE gel and optimize transfer conditions for high-molecular-weight proteins.
- In FC and IF/ICC, the antibody can be used to detect CD101 on T-cell subsets, dendritic cells, or leukemic blasts; validate in the relevant sample system and consider Fc blocking steps.
- Cross-reactivity with mouse allows translational applications in mouse models; however, verify reactivity and specificity in your model system through positive and negative controls.
