- CD101, also known as IGSF2, V7, and EWI-101, is an immunoglobulin superfamily member that functions as a cell surface glycoprotein.
- It acts as an inhibitor of T-cell proliferation induced by CD3, suppressing IL2RA expression and IL2 secretion.
- CD101 inhibits tyrosine kinases required for IL2 production and cellular proliferation, and blocks phospholipase C-gamma-1 phosphorylation and subsequent intracellular calcium flux.
- It prevents nuclear translocation of nuclear factor of activated T-cell (NFAT) to the nucleus, thereby modulating T-cell activation.
- CD101 plays a role in T-cell inhibition via IL10 secretion by cutaneous dendritic cells and may serve as a marker for CD4+ CD56+ leukemic tumor cells.
- Subcellular localization is at the membrane; the protein contains immunoglobulin domains, disulfide bonds, and glycosylation sites.
- Tissue expression includes lung, thymus, and small intestine; it is detected on activated T-cells, monocytes, granulocytes, dendritic cells, and some leukemic cells.
