Biological Background: CD101 Function and Localization
- CD101, also known as immunoglobulin superfamily member 2 (IGSF2), V7, or EWI-101, is a transmembrane glycoprotein belonging to the immunoglobulin superfamily.
- It functions as a negative regulator of T-cell proliferation: it inhibits CD3-induced T-cell activation by blocking IL2RA expression, IL2 secretion, and intracellular calcium mobilization.
- CD101 suppresses phospholipase C-gamma-1 phosphorylation and prevents nuclear translocation of NFAT, thereby limiting IL2 production.
- In cutaneous dendritic cells, CD101 contributes to T-cell inhibition through IL10 secretion.
- Expressed on the plasma membrane, CD101 is found in lung, thymus, small intestine, and on immune cells including activated T-cells, monocytes, granulocytes, and dendritic cells.
- It is a recognized marker for CD4+ CD56+ leukemic tumor cells and is present on Langerhans cells from Langerhans cell histiocytosis patients.
- Contains immunoglobulin-like domains and a signal peptide, with a predicted molecular weight of ~115 kDa.

- The immunogen (aa 21-410) covers the extracellular region; the antibody may recognize native and denatured CD101. Consider testing under non-reducing and reducing conditions for Western blot.

- Activation of T-cells (e.g., with CD3/CD28) can upregulate CD101 expression; include appropriate controls and stimulation protocols to enhance detection.
