Biological Background: CD101 Function and Localization

  • CD101, also known as Immunoglobulin superfamily member 2 (IGSF2), cell surface glycoprotein V7, or EWI-101, is a transmembrane glycoprotein containing immunoglobulin domains.
  • It functions as a negative regulator of T-cell proliferation induced by CD3, inhibiting IL2 receptor alpha (IL2RA) expression and IL2 secretion.
  • CD101 suppresses key signaling events including tyrosine kinase activation, phospholipase C-gamma-1 (PLCG1) phosphorylation, and intracellular calcium mobilization upon CD3 engagement, preventing nuclear translocation of nuclear factor of activated T-cells (NFAT).
  • This protein is also involved in T-cell inhibition through IL-10 secretion by cutaneous dendritic cells, suggesting a role in immune tolerance.
  • CD101 is expressed in lung, thymus, small intestine, and immune cells such as cutaneous dendritic cells, activated T-cells, monocytes, and granulocytes, as well as in epithelial cells with dendritic morphology.
  • It is notably expressed in a subset of leukemic cells, particularly CD4+ CD56+ blastic tumor cells and Langerhans cells from Langerhans cell histiocytosis, serving as a potential disease marker.

Experimental Guidance and Technical Tips

  • The immunogen encompasses amino acids 21–410 of human CD101, covering most of the extracellular region; this polyclonal antibody may recognize multiple epitopes within this domain.
  • For Western blotting, a predominant band at approximately 115 kDa is expected, consistent with the calculated molecular weight of CD101 (UniProt: 115,109 Da); equivalent loading controls and molecular weight markers are recommended.
  • In ELISA, the recombinant immunogen can serve as a positive control to optimize coating conditions and to establish assay linearity and sensitivity.
  • Since cross-reactivity is verified for human only, this antibody is suited for human samples; reactivity with other species should be assessed experimentally if needed.
  • Titrate the antibody empirically for each application and sample type, as polyclonal antibodies may exhibit lot-to-lot variation.
