Biological Background: Neprilysin Function and Localization
- Neprilysin (CD10) is a zinc-dependent metalloprotease belonging to the thermolysin-like family, exhibiting specificity for polypeptides of up to 30 amino acids (By similarity).
- Plays a critical role in the inactivation of opioid peptides such as Met- and Leu-enkephalins by cleaving a Gly-Phe bond (By similarity).
- Catalyzes the hydrolysis of several regulatory peptides, including bradykinin, substance P, and neurotensin (By similarity).
- Cleaves angiotensin-1, angiotensin-2, and angiotensin 1-9, contributing to the regulation of the renin-angiotensin system (By similarity).
- Involved in the degradation of atrial natriuretic factor (ANF), thereby modulating cardiovascular homeostasis (By similarity).
- Exhibits UV-inducible elastase activity toward skin preelastic and elastic fibers, linking it to skin extracellular matrix remodeling (PubMed:20876573). Related references: PMID:20876573
- Localizes primarily to the cell membrane as a type II integral membrane protein (signal-anchor).
- Post-translational modifications include glycosylation, disulfide bond formation, phosphorylation, myristoylation, and anchoring via a transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 52-750 of mouse CD10, representing the large extracellular domain; therefore, the antibody is expected to recognize native cell surface CD10. For flow cytometry, consider using live, non-permeabilized cells to detect surface expression.
- For Western blotting, the predicted protein weight is 86 kDa; however, due to glycosylation, the observed band may appear slightly larger. Validate the band specificity in your target tissue or cell lysate.
- In ELISA applications, use an appropriate recombinant or native CD10 protein as a standard to optimize assay conditions.
- Cross-reactivity is validated for mouse; if using in other species, verify sequence homology and perform pilot experiments.
