Biological Background: CCR6 Function and Localization

CCR6 (C-C chemokine receptor type 6) functions as a receptor for the chemokine CCL20, triggering intracellular calcium increase and cAMP signaling (PubMed:9169459, PubMed:20068036). Additionally, it binds beta-defensins such as DEFB1, DEFB4, and DEFB4A/B to mediate chemotaxis and bactericidal activity (PubMed:25585877, PubMed:25122636). As a seven-transmembrane GPCR, CCR6 localizes to the cell membrane and contains conserved disulfide bonds and glycosylation sites. It is predominantly found in spleen, lymph nodes, appendix, fetal liver, and on lymphocytes, T-cells, and B-cells, but not on granulocytes or monocytes.
Experimental Guidance and Technical Tips
- The immunogen was designed as a synthetic peptide corresponding to the C-terminal region (amino acids 300 to end) of human CCR6. This region is intracellular, so the antibody is likely to detect total CCR6 in denaturing Western blot conditions. For ELISA, consider using the immunizing peptide as a positive control to assess specificity.
- The predicted molecular weight of CCR6 is approximately 42 kDa. In Western blot experiments, a band near this size is expected; however, post-translational modifications (e.g., glycosylation) may result in higher apparent molecular weight.
- This antibody has been tested in WB and ELISA with human samples. For other species, cross-reactivity may be limited; validate in the relevant sample system.
- For immunohistochemistry or flow cytometry, note that the antibody may not recognize native epitope due to the peptide immunogen; empirical optimization is advised.
