Biological Background: CCR2 Function and Localization
- C-C chemokine receptor type 2 (CCR2), also known as Monocyte chemoattractant protein 1 receptor (MCP-1-R), is a G protein-coupled receptor (GPCR) encoded by the CCR2 gene. Alternative splicing generates isoforms including CCR2A and CCR2B.
- Functions as a key receptor for chemokines CCL2 (MCP-1), CCL7, and CCL12, and transduces signals from CCL13. Upon ligand binding, it mediates monocyte/macrophage chemotaxis and migration through activation of the PI3K cascade, Rac signaling, and lamellipodium formation. - Also serves as a receptor for the beta-defensin DEFB106A/DEFB106B, highlighting a role in innate immunity. Related references: PMID:23938203
- Acts as an alternative coreceptor with CD4 for HIV-1 infection, facilitating viral entry into cells.

Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide spanning the N-terminal region (aa 1-100) of human CCR2, which may confer epitope specificity. For applications requiring native conformation, validate the antibody under non-denaturing conditions if needed.
- In Western blotting, CCR2 is predicted to migrate at ~42 kDa; however, post-translational modifications such as N-linked glycosylation and phosphorylation can cause heterogeneity. Optimize sample preparation and gel conditions appropriately.
- For ELISA, consider pairing this monoclonal antibody with a capture/detection antibody recognizing a distinct epitope to enhance assay sensitivity. Validate performance with relevant recombinant proteins or cell lysates.
- CCR2 is expressed on monocytes and certain immune cells; therefore, suitable positive controls include peripheral blood mononuclear cells (PBMCs), monocytic cell lines (e.g., THP-1), or tissues rich in macrophages. Always include negative controls to confirm specificity.
