Biological Background: CCR2 Function and Localization
- CCR2 is a receptor for CCL2/MCP-1, CCL7, CCL12, and CCL13, mediating monocyte and macrophage chemotaxis via PI3K, Rac, and lamellipodium protrusion. It also functions as a receptor for DEFB106A/DEFB106B. Related references: PMID:23408426 PMID:38157855 PMID:8048929 PMID:23938203
- CCR2 is highly expressed on classical CD14+/CD16- monocytes and weakly on CD14+/CD16+ monocytes, type 2 DCs, and plasmacytoid DCs, with expression also detected on IL2-activated NK cells. Related references: PMID:9058802 PMID:38157855
- The protein is an integral membrane protein localized to the cell membrane, characterized by seven transmembrane domains typical of GPCRs.
- CCR2 undergoes several post-translational modifications, including disulfide bond formation, glycosylation, phosphorylation, and sulfation, which may regulate its function.
- It promotes Th17 differentiation during inflammation and facilitates thymocyte egress by enhancing S1P1R expression via FOXO1 signaling (by similarity).
- In the nervous system, CCR2 contributes to peripheral nerve injury-induced neuropathic pain and enhances NMDA receptor-mediated synaptic transmission via MAPK/ERK (by similarity). It also mediates macrophage recruitment to the brain after injury (by similarity).
- Alternative splicing generates isoforms such as CCR2A and CCR2B, and the protein is implicated in HIV-1 infection as a co-receptor with CD4.
Experimental Guidance and Technical Tips
- This polyclonal antibody, generated against a synthetic peptide spanning amino acids 150-250 of human CCR2, is designed for Western blot (WB) and ELISA applications. Its cross-reactivity with mouse suggests conserved epitopes, making it suitable for comparative studies.
- For WB, consider using whole-cell lysates from human or mouse monocyte/macrophage cell lines, and validate the observed band size (approximately 42 kDa) with appropriate positive and negative controls.
- In ELISA, pairing this antibody as a capture or detection reagent may be optimized by testing serial dilutions and confirming specificity with recombinant CCR2 protein.
- Given the transmembrane nature of CCR2, sample preparation with mild detergents could help preserve conformation for detection.
- Always titrate the antibody in your own experimental system to determine optimal working concentrations.
