Biological Background: CCL8/MCP-2 Function and Localization
- CCL8, also known as MCP-2 (monocyte chemoattractant protein 2), is a member of the CC chemokine family and is encoded by the CCL8 gene in humans.
- It is a secreted chemotactic cytokine that attracts monocytes, lymphocytes, basophils, and eosinophils, playing a role in inflammatory responses and neoplasia.
- The mature protein is processed from a signal peptide-containing precursor; an alternative processed form, MCP-2(6-76), lacks monocyte chemotactic activity but can inhibit the chemotactic effects of CCL7, CCL2, CCL5, and CCL8 itself.
- CCL8 binds heparin, a property that may localize it to endothelial surfaces and extracellular matrix to establish chemokine gradients.
- Subcellularly, it is secreted into the extracellular space.
- Expression is highest in the small intestine and peripheral blood cells, with intermediate levels in heart, placenta, lung, skeletal muscle, thymus, colon, ovary, spinal cord, and pancreas, and lower in brain, liver, spleen, and prostate.
