Biological Background: CCL5/RANTES Function and Localization
- CCL5 (C-C motif chemokine 5), also known as RANTES, EoCP, SIS-delta, and TCP228, is a secreted pro-inflammatory chemokine encoded by the CCL5 gene (SCYA5).
- It exerts chemoattractant activity toward blood monocytes, memory T-helper cells, and eosinophils, and triggers histamine release from basophils, leading to eosinophil activation. It engages chemokine receptors including CCR1, CCR3, CCR4, and CCR5. Related references: PMID:1380064, PMID:15923218, PMID:16791620
- CCL5 is a major HIV-suppressive factor released by CD8+ T-cells; recombinant RANTES inhibits diverse strains of HIV-1, HIV-2, and SIV in a dose-dependent manner. Related references: PMID:1380064, PMID:15923218, PMID:16791620
- Two naturally processed forms, RANTES(3-68) and RANTES(4-68), display altered chemotactic and HIV-suppressive potencies; RANTES(3-68) is a more potent inhibitor and a natural chemotaxis antagonist. Related references: PMID:1380064, PMID:15923218, PMID:16791620
- Beyond chemotaxis, CCL5 acts as an agonist of GPR75, stimulating inositol trisphosphate production and calcium mobilization, and modulates signaling pathways linked to neuron survival and insulin secretion. Related references: PMID:23979485
- Expressed predominantly in T-cells and macrophages, and detected in follicular fluid at the protein level.
- Post-translational modifications include disulfide bond formation, glycosylation, and oxidation; the protein is secreted and serves as a key mediator in inflammation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length mature CCL5 protein (residues 1–91), enabling recognition of both native and denatured forms. This supports WB and ELISA applications.
- For Western blot, a starting dilution of 1:500–1:2,000 is recommended; optimize empirically with your sample type and detection system.
- In ELISA, the antibody can be used as a capture or detection reagent; pairing with a monoclonal CCL5 antibody may improve assay performance in sandwich formats.
- Validate specificity using relevant human cell models (e.g., stimulated T-lymphocytes, macrophages) or body fluids known to contain CCL5.
