Biological Background: CCL4 Function and Localization

  • CCL4, also known as MIP-1β, ACT-2, LAG-1, and SCYA4, is a member of the CC chemokine family with a mature mass of ~10 kDa after signal peptide cleavage.
  • It is a secreted monokine with inflammatory and chemokinetic properties, acting as a chemoattractant for various immune cells.
  • CCL4 binds to the chemokine receptor CCR5, and is recognized as one of the major HIV-suppressive factors produced by CD8+ T-cells.
  • Recombinant CCL4 dose-dependently inhibits infection by diverse strains of HIV-1, HIV-2, and SIV.
  • The N-terminally processed form MIP-1β(3-69) retains the ability to down-modulate CCR5 surface expression and block CCR5-mediated HIV-1 entry, and also acts as a ligand for CCR1 and CCR2 isoform B.
  • The protein contains conserved disulfide bonds critical for its chemokine structure and function, and is identified as a secreted cytokine involved in chemotaxis and inflammatory response.
