Biological Background: CCL22 Function and Localization
- CCL22, also known as macrophage-derived chemokine (MDC) and stimulated T-cell chemotactic protein 1 (STCP-1), is a secreted CC chemokine involved in immune cell trafficking.
- It plays a role in the recruitment of activated/effector T-lymphocytes to inflammatory sites and is chemotactic for monocytes, dendritic cells, and natural killer cells.
- Acts as a mild chemoattractant for primary activated T-lymphocytes and a potent chemoattractant for chronically activated T-lymphocytes; it has no chemoattractant activity for neutrophils, eosinophils, or resting T-lymphocytes.
- Binds to the CCR4 receptor; processed forms MDC(3-69), MDC(5-69), and MDC(7-69) appear to be inactive.
- Highly expressed in macrophages, monocyte-derived dendritic cells, and thymus; also found in lymph node, appendix, and skin in inflammatory conditions such as atopic dermatitis and psoriasis.
- Subcellular localization is predominantly secreted, consistent with its role as a cytokine.
- Keywords: chemotaxis, cytokine, disulfide bond, secreted, signal.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 25-93 of human CCL22, covering the mature secreted form. Consider the antibody primarily for IHC-P and ELISA as indicated, and validate in the relevant sample system.
- Due to the polyclonal nature, cross-reactivity with rat is listed; verify performance in rat-derived samples if required.
- As a secreted protein, optimize fixation and permeabilization protocols for IHC-P detection; include appropriate positive controls such as inflamed lymph node or skin tissue.
- For ELISA, consider using recombinant CCL22 as a standard and confirm linearity and sensitivity in the matrix of interest.
